Discovery of new white blood cell reveals target for better vaccine design

ScienceDaily (July 27, 2012) ? Researchers in Newcastle and Singapore have identified a new type of white blood cell which activates a killing immune response to an external source -- providing a new potential target for vaccines for conditions such as cancer or Hepatitis B.

Publishing in the journal Immunity, the team of researchers from Newcastle University in collaboration with A*STAR's Singapore Immunology Network (SIgN) describe a new human tissue dendritic cell with cross-presenting function.

Dendritic cells (DCs) are a type of white blood cell that orchestrate our body's immune responses to infectious agents such as bacteria and viruses, as well as cancer cells. They are also very important for eliciting the immune response generated by vaccines.

DCs kick start an immune response by presenting small fragments of molecules from micro-organisms such as bacteria and viruses, or from vaccines or tumours, called antigens on their surface. This leads to activation of another white blood cell subset called T cells, which specialise in killing cells and are crucial for eliminating cancerous or infected cells. Most cells are only able to present antigens from within themselves, and so will only elicit an immune response if they are infected themselves. Only a specialised subset of DCs is able to generate a response to an external source of antigen, for example bacteria, vaccines and tumours.

The identity of human tissue DCs that are capable of presenting external antigen to activate the cell-killing response by T cells -- a process termed 'cross-presentation' -- has remained a mystery. Their discovery, as revealed by this research, will help scientists to design better targeted vaccine strategies to treat cancer and infections such as Hepatitis B.

"These are the cells we need to be targeting for anti-cancer vaccines," said Dr Muzlifah Haniffa, a Wellcome Trust Intermediate Fellow and Senior Clinical Lecturer at Newcastle University. "Our discovery offers an accessible, easily targetable system which makes the most of the natural ability of the cell." The researchers also showed for the first time that dendritic cell subsets are conserved between species and have in effect created a map, facilitating the translation of mouse studies to the human immune system.

"The cross-species map is in effect a Rosetta stone that deciphers the language of mouse into human," explains Matthew Collin, Professor of Haematology from Newcastle University.

In the paper the researchers describe how the cross-presenting DCs were first isolated from surplus plastic surgery skin which was digested to melt the gelatinous collagen to isolate the cells. This research will have significant impact on the design of vaccines and other targeted immunotherapies.

The Rosetta Stone of our immune system: Mapping Human and Mouse dendritic cells

The Newcastle University team in collaboration with A*STAR's Singapore Immunology Network (SIgN) have for the first time ever aligned the dendritic cell subsets between mouse and humans allowing the accurate translation of mouse studies into the human model for the first time.

The researchers isolated the dendritic cells from human blood and skin and those from mouse blood, lung and liver. Using gene expression analysis, they identified gene signatures for each human dendritic cell subset. Mouse orthologues of these genes were identified and a computational analysis was performed to match subsets across species.

This provides scientists for the first time with an accurate model to compare DCs between species. Professor Matthew Collin explains: "This is in effect a Rosetta stone that deciphers the language of mouse into human. It can put into context the findings from the extensive literature using mouse models to the human settings."

Dr. Haniffa added: "These gene signatures are available in a public repository accessible for all researchers to benefit from the data. It will allow detailed knowledge of individual human dendritic cell subsets to enable specific targeting of these cells for therapeutic strategy."

Romney: ?No option should be excluded? in stopping a nuclear Iran

Romney in Jerusalem (Uriel Sinai/Getty Images)

JERUSALEM?Speaking against the backdrop of the historic Old City, Mitt Romney signaled he supports Israel's right to "defend itself" against its enemies and said "any and all measures" should be considered in the world's efforts to stop Iran from developing a nuclear weapon.

The Republican candidate said preventing a nuclear Iran should be America's "highest national security priority." And while he argued that diplomatic efforts should continue, Romney insisted that all options should be on the table in dealing with Iran, including a military strike.

"We should employ any and all measures to dissuade the Iranian regime from its nuclear course, and it is our fervent hope that diplomatic and economic measures will do so," Romney said in an outdoor speech at Mishkenot Sha'anamim, a cultural center in Jerusalem. "In the final analysis, of course, no option should be excluded. We recognize Israel's right to defend itself, and that it is right for America to stand with you. "

Romney pushed back against criticism that those arguing in favor of leaving the military option open are "reckless and provocative and inviting war." "The opposite is true. We are the true peacemakers," Romney said.

He argued that the threat of military action would put more pressure on the ayatollahs in Iran, whom he accused of "testing our moral defenses."

"They want to know who will object and who will look the other way," Romney said. "My message to the people of Israel and the leaders of Iran is one and the same: I will not look away, and neither will my country."

In a message sure to appeal to Jewish voters, with whom Republicans are aiming to make inroads ahead of November's election, Romney repeatedly emphasized the "shared values" and "shared interests" between the United States and Israel.

Romney avoided directly criticizing President Barack Obama but slipped in a dig at his Democratic rival, arguing that public differences between the U.S. and Israel only embolden Israel's enemies.

"We cannot stand silent as those who seek to undermine Israel, voice their criticisms.? And we certainly should not join in that criticism," Romney said. "Diplomatic distance in public between our nations emboldens Israel's adversaries.

By history and by conviction, our two countries are bound together.?No individual, no nation, no world organization, will pry us apart. And as long as we stay together and stand together, there is no threat we cannot overcome and very little that we cannot achieve."

Romney is dining with Israeli Prime Minister Benjamin Netanyahu on Sunday night. On Monday, he'll headline a major fundraiser for his campaign among top Jewish supporters, many of whom were in the audience for Sunday's speech. Among those in attendance: Las Vegas casino magnate Sheldon Adelson, who has contributed millions of dollars to a pro-Romney super PAC.

On Monday afternoon, Romney heads to Poland, the final stop on his weeklong overseas tour.

Writing Kindle Ebooks Part 7 ? Publishing Your Kindle Book on ...

By: Joy Burgess

Previously, we?ve discussed different aspects of writing Kindle eBooks. In the last post, ?Writing Kindle Ebooks Part 6,? we talked about formatting your book to get it ready for publishing. Now that your book is well written and formatted, you?re ready to publish it on Amazon.com. Whether you?re publishing on Amazon for the first time or you just need a refresher course, the following information will help you through the publishing process.

Two Uploading Options to Consider

By now, your eBook is complete and formatted, whether you wrote it on your own or hired Kindle book writers to take care of that step for you. Before uploading the eBook, you have two different uploading options to consider. When you start the publishing process, you?ll have to choose whether to go the KDP Select (Kindle Direct Publishing) route or to skip this option.

What is KDP Select? This program locks in your eBook exclusively to Amazon for 90 days. During this time, you?ll be given five promotional days free. On these days, you can give the eBook away free, but the book goes back on the market at a price after those days. Also, your eBook is made available in the Kindle Owners? Library for those 90 days as well.

The Kindle Owners? Library is a special option exclusively for Amazon Prime Members. Members have the ability to borrow an eBook for a month without due dates. However, they can only borrow these books on Kindle devices. The good news is that while members can borrow your book free of charge, you still earn money. This is only an option if you decide to enroll in the KDP Select program.

Certain restrictions do apply if you decide to enroll in KDP Select. The following are a few of the restrictions you?ll have to follow:

-????????? The book can only be offered free for five days during the 90-day period

-????????? You may not post your book on your blog

-????????? The book cannot be offered as an opt-in bonus

-????????? During the 90 days, you may not distribute the book anywhere else

-????????? The book cannot be offered to iBooks or Nook during this time

-????????? You have the ability to opt-out at any time, but you still have to follow the restrictions until the 90 days are up

-????????? If you continue to opt-in, your book will renew for another 90 days, also providing you with five more free days of promotion

If you choose not to go the KDP Select route, you?re still able to upload the eBook to Amazon Kindle. However, it won?t be included in the Kindle Owners? Library

When making your choice, keep in mind that not every reader has a Kindle. However, readers can download the Kindle App free and it is available for many systems, including the following:

-????????? Mac

-????????? Blackberry

-????????? Windows PC

-????????? iPhone

-????????? Android

-????????? iPad

-????????? Windows 7 Phones

The Publishing Process ? Step-By-Step Guide

After deciding on the uploading option you want to use, you?re ready to start the publishing process. Before you start, make sure you already have your full-length eBook saved in Microsoft Word and the image file saved that you plan to use for your cover.

Step #1 ? Save Your eBook as Filtered HTML

Once you have your Word document open, you need to save the document as filtered HTML. This prepares your file for upload to Amazon?s website.

Step #2 ? Go to Amazon?s Self-Publishing Page

Go to Amazon?s Self-Publishing Page and sign in to your Amazon account. If you don?t have an account, sign up for one. Once logged in, you?ll be at the Amazon Kindle Author Bookshelf, where you?ll want to click on the button that says, ?Add a New Title.?

Step #3 ? Enter Important Book Information

Now you?ll have a page in front of you where you can enter the details for your book. Add the title. If you are writing a series, use the series option to input series information. Then, you?ll need to enter a description of your eBook. Keep in mind that this description is extremely important, since it?s what readers will see when viewing your title.

Step #4 ? Add Contributors

After entering the description of your Kindle book, you?ll notice an ?Add Contributors? button. This is where you?ll provide author information. If others have contributed to the book, such as editors or illustrators, their information should go here as well. After adding contributors, you can select your language, the publication date, publisher information (if any) and an ISBN number if you have one. Keep in mind, you are NOT required to have an ISBN number when publishing with Kindle.

Step #5 ? Verification of Publishing Rights

Now you?ll be asked for verification of publishing rights. More than likely, you?ll be choosing the second choice offered, which states, ?This is not a public domain work and I hold the necessary publishing rights.?

Step #6 ? Add Keywords and Categories

To target potential customers, you?ll need to choose a category for your book and add relevant keywords in the ?search keywords? box. Keep in mind, those keywords are important, since this will help readers find your book when searching for specific information.

Step #7 ? Upload the Cover

Now you?ll be given the option to upload your cover for the book. While this is optional, a good cover is important, since the cover shows up in Kindle libraries and on book detail pages. When deciding on a cover image, something that looks great as a thumbnail image and as a full sized image is important.

Step #8 ? Upload the eBook File

Before unloading the book file, you?ll be asked about digital rights management. It?s usually a good idea to choose the option ?enable digital rights management,? which helps protect the rights of your book. Then you can browse for the book file and once you select the Filtered HTML file you saved earlier, you can hit the ?Upload? button. At this point, you can save the book as a draft or you can save it and continue to last few steps.

Step #9 ? Handle Royalty Information

The last few steps have to do with your royalties for the eBook. Unless you only have rights to publish your manuscript in a particular territory, you may choose ?Worldwide rights.?

Then you?ll be given royalty options to choose from. You can choose the 70% royalty rate, which comes with a small delivery cost, or you can choose the 35% royalty rate, which has no cost for delivery. Most people find that the 70% royalty rate offers them the most profit. You can also choose to have UK and DE prices set automatically based upon the price you set for the U.S.

Step #10 ? Decide on Kindle Book Lending

Enabling Kindle Book Lending means that Kindle users have the option to share your eBook with other users. While many may not like this option, it can be helpful for first time authors, since word-of-mouth can be a powerful marketing tool.

Step #11 ? Save and Publish

You may want to take the time to review the Terms and Conditions, which is right near the checkbox you must click before saving and publishing your Kindle book. If you?re comfortable with those terms and conditions, you can check the box to note that you understand these terms. Once you?ve checked the box, you can hit ?Save and Publish,? which will complete your upload.

At this point, it may take some time for Amazon to get your eBook into the Kindle Store. While you can easily edit, delete and un-publish the book in the Amazon Kindle Author Bookshelf, you?ll probably want to allow Amazon to get your book up before making changes. When the book is ready and available, it?s time to start promoting the eBook, which is something we?ll discuss in the next post.

Amazon does make it easy for anyone to self-publish an eBook. Take advantage of what Kindle eBooks have to offer and get your own book out there today. If you haven?t started to write Kindle books, you do have the option of hiring Kindle book ghost writers to do the job for you.

Stay tuned for the last post in the series, ?Writing Kindle eBooks,? which will offer you helpful tips and advice on promoting and marketing your eBook for the great selling results.

(AP) ? Fresh from her star turn as the latest Bond Girl, Queen Elizabeth II returned to Olympic Park for an encore Saturday, while the usually biting British press gave a resounding review: We are amused.

The queen visited with fawning British Olympians in the athletes village and rode to the top of the 377-foot (115-meter) Orbit tower beside the stadium, where on Friday night she officially opened the 2012 Games. Her husband, Prince Philip, and London Mayor Boris Johnson accompanied her.

Dressed in a royal blue silk dress, crepe coat and matching hat, the queen could be heard remarking at the views of the London skyline and countryside. She wore a brooch given to her in 1948 ? the last time London hosted the Olympics and four years before her accession to the throne.

In describing her role in the opening ceremony, created by director Danny Boyle, Johnson said the queen had told him she was "very, very impressed with the success of her first film appearance, her first dramatic venture. It was very funny and seems to have gone down particularly well with the international audiences."

And, it seems, with the British media.

The Guardian called the queen's appearance "one of the highlights of Danny Boyle's tumultuously inventive opening ceremony of the 2012 Games. ... Now, thanks to Boyle, we really have seen everything."

"I was worried that there was too much self-parody, that the world might be laughing at us," wrote columnist Giles Coren in The Times of London. "But they were laughing with us. They were silently awed."

The show-stopping moment featured the queen making her acting debut alongside Daniel Craig, who portrayed James Bond in a film beamed into the stadium.

Boyle's production showed Bond striding into Buckingham Palace to escort his VIP guest to the Olympic ceremony. Many had expected a famous actress to play the role of the queen, and there was an audible gasped as the real-life royal was seen to swivel round in her desk chair and declare: "Good evening, Mr. Bond."

Buckingham Palace confirmed that the 86-year-old monarch agreed last year to participate. The filming took place at the palace in March and April and featured two of the queen's corgi dogs: Monty and Holly.

At the end of the film segment, two stuntmen dressed as Bond and the queen parachuted from a helicopter into the stadium, and moments later Elizabeth and Philip emerged in the stands.

Boyle told NBC television that the queen had been a natural. "You don't have to tell her something twice," he said. "She picks it up straight away, about cameras and angles."

The sequence has already provided the defying images of the games, according to the Daily Telegraph in Australia, where the queen also reigns.

"A few hundred years ago director Danny Boyle could have been sent to the Tower for even suggesting such treason," the Sydney newspaper said. "But as if to show how far England and the monarchy have come in that time, Her Majesty not only let Boyle get away with it. She was actually in on the joke."

The German newspaper Die Welt wrote that "often seen as reserved and unapproachable, the queen changed all that alongside James Bond."

The queen offered more surprises Saturday as she met about 150 members of the British Olympic team. Spectators took in the scene from balconies draped in Union Jack flags.

"For her to come through and meet the athletes, and see where we're living, it was amazing," said Rose Anderson, 24, a member of the women's basketball team.

"She went inside one of the athletes' bedrooms and chatted to us. It was just awesome, especially after last night," she said. "Last night will never be topped. I'll never get an experience like that again, and then this will never be topped either."

The queen also met athletes from other nations in the dining hall.

"She's beautiful," said Jess Fox, who competes for Australia in the canoe slalom. "We saw her from afar last night in the stadium, but it was amazing to see her like this."

Children's Health & Fitness ? Magnetic Fields And Childhood Obesity

From The Science Blog?..

In-utero exposure to relatively high magnetic field levels was associated with a 69 percent increased risk of being obese or overweight during childhood compared to lower in-utero magnetic field levels, according to a Kaiser Permanente study that appears in the current online version of Nature?s Scientific Reports.

Researchers conducted the prospective cohort study, in which participating women in Kaiser Permanente?s Northern California region carried a meter measuring magnetic field levels during pregnancy and 733 of their children were followed up to 13 years, to collect clinically recorded information on growth patterns. On average, 33 weight measurements per child were collected.

Researchers noted a dose response relationship with increasing in-utero magnetic field levels being associated with further increased risk of obesity or being overweight. The observed association and supporting evidence provide the first epidemiologic findings that link increasing exposure to environmental magnetic fields, especially in-utero exposure, over the last few decades with the rapid rise in childhood obesity during the corresponding decades, according to the authors.

Willard says he did nothing to warrant arrest

(AP) ? Fred Willard said his recent lewd conduct arrest at an adult movie theater was "very embarrassing" but insisted he did nothing wrong.

"It's the last time I'm going to listen to my wife when she says, 'Why don't you go and see a movie?'" Willard said during an appearance Thursday on Jimmy Fallon's NBC show "Late Night."

The 72-year-old actor and Fallon cracked racy jokes about possible titles for the movie that might have been playing when Willard was arrested July 18 during a routine check by police at the Hollywood-area theater.

Then the conversation turned serious for Willard, whose credits include the movie "Best in Show" and guest roles on "Modern Family" and other TV shows.

"Fred, I know this must be a tough time," host Fallon asked.

"It's very embarrassing. It's as embarrassing as hell," Willard replied. "But let me say this: Nothing happened. I did nothing wrong. Everything is being sorted out as we speak."

He has great respect for the police, Willard added, "but if you're in the wrong place at the wrong time, everything seems suspicious."

He's tried to laugh it off but the incident is "very painful," said Willard, who was fired as narrator of the new PBS series "Market Warriors" immediately after his arrest.

The Los Angeles City Attorney's office determined that Willard's case was eligible for a diversion program that will keep him from being formally charged with lewd conduct if he completes the required courses. Willard will pay $380 for the program.

Talk shows can be useful for celebrity efforts at damage control. In 1995, Hugh Grant visited Jay Leno on NBC's "Tonight" after being arrested with a prostitute on Hollywood's Sunset Strip.

Fallon was sympathetic to Willard.

"You're a good man. ... You're one of my favorites," Fallon said, then needled him with, "You can come to our show and do whatever the heck you want, except for certain things."

Willard was nominated four times for Emmys for guest roles on "Modern Family" and "Everybody Loves Raymond." In Pixar's 2008 hit "WALL-E," he voiced the character of Shelby Forthright, the CEO of a ubiquitous big-box chain called Buy'n'Large.

In addition to "Best in Show," Willard has also appeared in other Christopher Guest mockumentary films, including "This is Spinal Tap" and "Waiting for Guffman."

Willard is hosting "Trust Us With Your Life," an improv series airing this summer on ABC.

EYES ON LONDON: Crowd in the mood, ceremony soon

LONDON (AP) ? Around the 2012 Olympics and its host city with journalists from The Associated Press bringing the flavor and details of the games to you:

___

THE ANTICIPATION

From AP's Fergus Bell in place and awaiting the opening ceremony:

"A Mexican wave just started. The crowd are getting fired up. The scene is set on the stage in front of me depicting a rural idyll. There are tractors and a water wheel. There is a small cottage in the middle with two smoking chimneys. I have seen similar scenes in the English countryside but this looks a little cleaner, a little more fairytale.

? Fergus Bell ? Twitter http://twitter.com/fergb

___

GETTING THERE

AP's Cassandra Vinograd reports from the streets ? well, the railway tracks ? of London:

This reporter was in a bit of a panic ? left work late and wondered how she would ever get through the crowds to board a train to Olympic Park.

But tubes were not crowded and St. Pancras International, where you catch the high speed Javelin train, is a breeze. There were no lines whatsoever to get on the Javelin ? despite crowd-flow measures (barricades, like what airports use for long check-in lines). The line spaces were all empty.

The only glitch was a few travelers grumbling over a broken ticket machine, leaving only one working dispenser on the Javelin platform.

But no worries in the end. With so few jostling for space, everyone got a ticket in time.

___

NO KNOCKOFFS

Egypt's Olympic team won't be wearing knockoffs after all at the London Games.

Nike announced Friday it was donating its gear to the Egyptian athletes. This after the country's committee chairman said they'd turned to a Chinese distributor because the real thing was just too expensive.

Not so during the opening ceremony, though. Nike's shipment isn't due to arrive in London until Saturday.

WHITE RIBBONS

The widow of Alexander Litvinenko, the exiled former Russian agent who died from a mysterious case of radiation poisoning, is calling on Londoners to protest Russian President Vladimir Putin's visit to the Olympics.

Litvinenko issued a statement shortly before his 2006 death, accusing Putin of ordering him to be poisoned. He had been living in London since fleeing Russia in 2000. In a book written in exile, he accused Russia's main security agency, for which he once worked, of staging the 1999 apartment building bombings that were blamed on Chechen rebels and used as a justification for starting the second Chechen war.

British-Russian relations suffered a serious setback in 2007 after Russia refused to extradite the man that Britain named as the main suspect in Litvinenko's death.

Marina Litvinenko on Friday issued a statement saying "Mr. Putin's agents killed my husband" and called on Londoners wo wear white ribbons, the emblem of the Russian opposition, to proest Putin's visit Aug. 2 that is to include watching the judo.

?Jim Heintz ? Twitter http://twitter.com/jeheintz

___

WHERE TO WATCH?

AP's Jenna Fryer reports from the streets of London:

Is there no such thing as a sports bar here?

Looking for a pub where I can hang with the locals and watch the opening ceremony is proving to be more challenging than I expected.

Since arriving, I've noticed that not that many pubs have televisions, so I've had to ask around for recommendations for the ceremonies. The first two I called ? one of which was the favorite watering hole of the late Amy Winehouse ? said they had no plans to show the ceremonies. "Tonight's just a regular pub night," I was told.

It seemed strange to see bars without TV's, but to not have such a big event blaring from every television in the country tonight is just bizarre.

PREPPED FOR HISTORY

Double-amputee runner Oscar Pistorius has arrived in London for his historic debut at the Olympics and says he's feeling "a mixture of pride, happiness and anticipation" at the end of his long journey to compete at the highest level in able-bodied competition.

Pistorius made the short flight across from his training camp in northern Italy and says he's "really excited" to attend the opening ceremony.

The South African has been battling for four years to reach the 2012 games. He was cleared to compete against able-bodied competitors on his carbon fiber blades in 2008, ran two qualifying times for London in the last 12 months, but missed out on the crucial third under his country's strict qualifying criteria.

But the selectors had a last-minute change of heart weeks before the games and picked him for the individual 400 meters as well as the 4x400 relay.

In an email to the AP, Pistorius says: "This feels like a long time coming and I am ready! This is going to be, I hope, an incredible few weeks of my life."

His first run at the Olympics will be in the 400 heats on Aug. 4. It'll be history.

DIPLOMATIC MARATHON

It sounds almost as exhausting as the sport.

Britain's foreign policy chief William Hague is carrying out a frantic round of Olympic diplomacy, greeting foreign dignitaries arriving for the Summer Games ? and taking a chance to press them over trade, the Middle East and the violence in Syria.

Already, Hague has met with the prime ministers of Haiti, Lebanon, Kosovo, Panama's president and U.N. Secretary-General Ban Ki-moon. In the coming days he has talks scheduled with VIPs from Bangladesh, India, Singapore and New Zealand.

___

LITTLE BOYS?

The British men's volleyball team is playing in its first-ever Olympics. But truth be told, if the men hadn't earned an automatic berth as the host nation, they likely wouldn't be there.

The team is currently ranked 92nd in the world, and admittedly are a long shot for the gold. "Everybody sees us as the little boys of the volleyball competition. I hope we surprise some people," coach Harry Brokking says.

? Anne Peterson ? Twitter http://twitter.com/anniempeterson

___

HIDDEN CEREMONY

Britain's women's footballers say they will wear their opening ceremony uniforms even though they'll be about 155 miles (250 kilometers) away in Wales.

But they'll have to draw the curtains ? at least until late at night.

They have a special room to watch the ceremony on television but they'll not be allowed to be seen in their uniforms until the British team is introduced for its parade at about 11 p.m. local time.

Coach Hope Powell says "the girls really just want to embrace it and be part of it" but admits: "It will be odd."

SUDAN DENIES ASYLUM REPORT

Here's a reaction just in from the Sudanese Embassy, denying reports that one of its Olympic runners has applied for asylum in the U.K.

"We herewith categorically refute allegations ... that a Sudanese member of those who qualified for the Olympic competition and have arrived in London is either missing (or) sought political asylum."

The male Olympian reportedly showed up Tuesday night at a police station near the northern English city of Leeds.

___

RIGHT IN THE MIDDLE

Draped in a British flag, 35-year-old Greg Kemp struck a pose below the Orbit, just outside Olympic Stadium shortly after entering the stadium.

The London native can't help but reflect on what his city has been through to get to this point.

"I remember winning the bid and the next day the bombings," Kemp says. "And from there to standing here and it's actually done, it's pretty spectacular.

"It's the journey of a city, isn't it? In 10 years' time, this is the history of London. And to make it to here, it's the center of the universe in my opinion and it's the greatest city in the world."

FOOD WITH NO NAME

Food choices are simple at the ExCel arena, the giant convention center hosting seven Olympic disciplines. Because McDonald's has exclusive rights to sell branded products, the dozen concessions at the venue are prohibited from using a specific name.

So, here's what's on offer on large signs:

?"Traditional Roast."

?"Sandwich."

?"Bagel"

?"Italian" or "Chinese."

? "Baked Potato" ? which, by the way, is often called a "jacket potato" in these parts.

CAGE FIGHTERS ... AND DOGS?

Timmy Duggan tried to come up with a way of describing what it's like to be a cyclist during a marquee event, and he left the rest of the American team in stitches.

"You have to have the endurance of a Siberian Husky for the five minutes of the race that truly matters," the first-time Olympian opined, "and it's the strength of an ultimate cage fighter, with fighter-pilot reactions, which is a pretty cool sport, when you think about it."

Duggan certainly won't be making any friends with mixed martial artists, though.

"We're pushing ourselves much harder than you even can in something like cage fighting," Duggan said. "We're going so hard that you can't stand up anymore. We just keep killing ourselves."

For the record, Duggan doesn't own a Siberian Husky, though he admitted that "I've read a lot of books about dog racing." And as for fighting, well ...

"I wouldn't take my chances," he said. "That's why I'm a bike racer."

KEEP DREAM-ING

You get the sense that LeBron James doesn't want to hear another word about it.

"Who's better ? 1992 or 2012?" That's been a talking point for weeks, the comparison between the 1992 Dream Team that went to Barcelona and this year's Olympic team.

"It's never going to happen," James said Friday of a hypothetical game between the two teams. "It doesn't matter how much conversation that we have about this team and the '92 Dream Team. It's never going to be played. Father Time is not going to allow that to happen. So it doesn't even matter."

That wasn't his way of downplaying expectations about what to expect from the U.S. squad in London.

Quite the contrary.

James, who won the NBA MVP award, the NBA Finals MVP award and led the Miami Heat to the title this past season, is fully expecting to add Olympic gold to his unbelievable year.

"I know one thing," James said. "I know we have a great team. And if we play as a team, like we're capable of doing, we can be one of the best teams ever assembled.

? Tim Reynolds ? Twitter http://www.twitter.com/ByTimReynolds

___

IT'S CALLED RACE WALKING

Of all the sports to watch at the Olympics, race walking is probably not No. 1 on many people's lists. But three-time NBA scoring champion Kevin Durant can't wait to check it out.

The Oklahoma Thunder star singled out the event Friday when asked what he plans to watch during his free time in London. That's when he's not trying to help Team USA win a second straight gold medal.

Mind you, he didn't get the name of the sport quite right.

"I want to watch some speed walking," he said, using the incorrect term for the event. "I didn't even know that was an Olympic sport."

TENNIS CHANGE

Update the roster at Wimbledon for Olympic tennis.

Laura Robson of Britain will play singles at the Olympics, replacing Petra Martic of Croatia after she pulled out because of injury.

Robson, a hometown fan favorite who is also playing doubles, will face Lucie Safarova of the Czech Republic in the first round. Robson won the Wimbledon junior title in 2008, and expectations have been high ever since. But she has won only one of her five matches in four main draw appearances.

The last British woman to win the Wimbledon titles was Virginia Wade in 1977.

AT OLYMPIC STADIUM

Olympic workers, concessionaires and opening ceremony participants are hustling around Olympic Stadium trying to put the final pieces in place for Friday night's opening ceremony.

Drinks are being transported, dance routines practiced and security workers are taking pains to keep passers by from entering the stadium while final preparations continue.

I did catch a quick glimpse before being turned away, seeing a stage with some sort of cottage as the centerpiece. Your guess is as good as mine.

Want To Improve Your Home? Some Smart Tips | Arty Apt

Don?t overspend your budget! When you set a budget, you?re ensuring that the home-improvement work you do is within a reasonable range for your home and neighborhood, as well as not putting you into the poor house. Create a comprehensive budget and then stick it through until the end of your project.

Before driving a nail or screw into a particularly stubborn piece of wood or other material, rub the screw with a bit of wax or a bar of soap. This lubricates the fastener and allows it to drive easily into the material without bending, slipping, or getting hung up.

You can add a room to your home without adding to its square footage. Use existing space to create that additional room by building a wall, which will turn one area into two. Closing off a partial wall or filling in an archway will allow you to realize that extra room.

You should always purchase the materials you need for home improvement projects in the largest possible lots. Building materials have some of the deepest volume discounts you will ever see. By planning ahead you can figure out how much material you are likely to need and buy it all at once. This will cost you much less than making multiple purchases.

Involving your children with your home improvement jobs can be fun, and will help you bond while building their self-esteem. Children are great helpers and enjoy feeling included, so giving them a small job to do makes them feel good about themselves, and this helps you at the same time.

Painting a room is perhaps the cheapest home improvement you can carry out. A fresh coat of paint in a different shade or texture can completely transform a room for minimal cost. If your walls are not in the best condition and you don?t have time to re-plaster them before painting, consider hanging wallpaper instead, to avoid drawing attention to the damage.

In a search for an simple home improvement, you found new kitchen hardware that you love but just doesn?t go with your existing style. Time to make some decisions. Where to start? Paint your existing cabinets, refinish them with stain, or buy new ones. Whatever you decide, make sure it?s in your home improvement budget.

If your chimney is dated back 100 years or more, you may want to consider replacing it. An old chimney becomes hazardous and decreases your homes efficiency as well. They can become cracked or clogged causing issues if you wish to use it, while more of your home?s heating and air can escape through an old chimney.

Resist the temptation to use fancy bricks with decorative faces in your next home improvement project. Not only is such brickwork an unnecessary expense, it is rarely as strong and durable as ordinary brick. Decorative bricks are easier to deface, spoiling their aesthetic advantages. Finally, decorative brick styles are rarely produced for long, making it incredibly difficult to find matching replacements for repair work later.

Make sure that you check your local building codes and get proper permits before you embark on any major remodeling project. Building codes and permits are meant to ensure that a specific building project adheres to safety standards. If a project does not follow building codes, there is a risk that the project may be unsafe.

A great home improvement method you can use is to actually check your faucets every year and do preventative maintenance on them. This way, you can avoid those annoying leaks and even those major breaks. It might seem like a hassle now, but you?ll be preventing a lot of work in the future.

As you can see, home improvement can actually be very satisfying. Learning how to do small jobs at your house fosters your sense of independence and boosts your confidence as a property owner. The do-it-yourself movement is absolutely within your reach after careful research, and reading this article is a great first step.

How to Choose a Good PPI Claims Service ? Payday Loans and ...

The hottest topic recently is about ppi claims. It concerns with someone who
pays policy to PPI without his consciousness based on the loan or credit agreement
and when he realizes on what he was done, he starts to make claims to reclaim the
PPI charges. There are many specialists who can help you in winning your claims but
among them you should be able to choose one that will give you their best service.
Some criteria should be owned by the specialist such as he should be able to find if
you have PPI, how many policies that you own, how many that you sold and how
much your owe is.
In 2011, there were millions of PPI policies that have been sold unfairly and
many banks that involve in certain loans companies were asked to pay more than
?1.9bn as the compensation to PPI. This fact is not fair for you as you are trapped in
such unfair situation. Here are some criteria of specialist in taking care of PPI claims
that you can consider as good advice. First, choose PPI claims service that employs
friendly staffs to work with you. It is important since they can look over your case
even it was made some years ago or even the agreement has been closed.
The second tip you can consider as a good advice is choosing a reliable PPI
claims service so you will not be tricked for a second time. Choose one that will help
you in taking your money back from the mortgage PPI. They should be able to give
your money back 100% and the fees they offer is reasonable for you. A good PPI
claims service is flexible and they can be asked to negotiate concerning the charge.
After all, if they cannot give your money back, they should be paid by you. They will
take the case only if they can handle your claims.
The third tip to consider is the PPI claims service should have good reliability
in front of their clients. You can find the best service online and compare one service
with others to choose the best one. Good services that are given to the clients can
be seen from the testimonials that are given by the clients to the PPI claims service.
If there are many positive reviews concerning their service you can choose them as
your partner.

ARLINGTON, Texas (AP) ? Derek Holland is intent on making up for the time he missed on the mound for the Texas Rangers, who keep winning series at home.

The left-hander certainly seems to be getting back into form after missing a month because of shoulder fatigue brought on when he lost about 15 pounds when he had a stomach virus.

Holland retired 20 of 22 batters over one stretch Wednesday night and struck out seven in 7 2-3 innings, and the Rangers scored the go-ahead run after a couple of wild throws by Josh Beckett in a 5-3 victory over the Boston Red Sox.

"I'm trying to do everything I can to step up and get those innings back, that I had lost in the month off," Holland said. "I put the pressure on myself. Obviously everybody else wants me to step things up, but I feel like I'm kind of doing it on my own."

The Rangers took two of three against Boston to win their seventh consecutive series at home. It came during a three-day span when they found out right-hander Colby Lewis was done for the season with a torn flexor tendon and Roy Oswalt skipped his scheduled start because of back stiffness.

In his fourth start since coming off the DL, Holland (7-5) allowed three runs and five hits with one walk.

"We needed exactly what he gave us," manager Ron Washington said. "He competed. He gave up two home runs, but he didn't let it affect him and kept getting after it."

Elvis Andrus was hit on the left forearm by a pitch to reach with one out in the seventh. He got to third on a single by Adrian Beltre and scored to break a 3-all tie when Beckett (5-9) threw a wild pitch that bounced several feet wide of the plate.

"Josh gave us a zero in the first inning, as promised, and pitched a good game," Boston manager Bobby Valentine said. "Nobody on in the seventh, then a wild pitch to score a go-ahead run. ... It's a damn shame."

Beltre was back in the lineup a day after being hit by a pitch thrown by Vicente Padilla that knocked off his helmet and sent him to the ground. Beltre, who insisted again before Wednesday's game that he didn't think that 0-2 pitch was purposely thrown at him, had a bruise but no concussion.

In what may have been a bit of retaliation with two outs in the first, Red Sox cleanup hitter Adrian Gonzalez was hit in the buttocks by a pitch. Home plate umpire Tim Tschida, the crew chief, pointed Gonzalez to first base and didn't issue any warnings.

There were no further issues, and it was clear by Beckett's reaction when he hit Andrus in the seventh that it was just a pitch that got away.

While retiring 20 of 22 batters, Holland gave up solo homers to Will Middlebrooks and Dustin Pedroia. He struck out the side in the seventh. The impressive stretch ended when Pedro Ciriaco had a two-out single in the eighth with Pedroia on deck.

Mike Adams retired Pedroia on a running catch by left fielder David Murphy. Joe Nathan worked a perfect ninth for his 20th save in 21 chances a day after allowing consecutive two-out walks and then giving up a run in a 2-1 loss to the Red Sox.

Beckett, who allowed nine hits and walked two in his seven innings, never appeared in the clubhouse when it was open to reporters after the game.

Jacoby Ellsbury drew a walk to start the game and eventually scored when Pedroia grounded into a double play.

Holland began his impressive streak after Middlebrooks led off the second with a single. Middlebrooks hit his 12th homer in the fourth, and Pedroia went deep in the sixth.

The Rangers scored three times in the fourth, when they got started with doubles by Michael Young and Murphy. It was Young's first extra-base hit in 11 games since the All-Star break, and Murphy's first RBI since June 28.

Murphy scored on Craig Gentry's perfectly executed suicide squeeze bunt to tie the game at 2. Ian Kinsler followed with an RBI bloop single that dropped after glancing off the heel of sliding right fielder Cody Ross' glove.

"We did some good execution," Washington said. "So it worked out for us."

Notes: Cruz was mired in a 5-for-33 slump with 11 strikeouts over 10 games before leading off the eight with a homer off Red Sox reliever Matt Albers. ... Cruz also had an impressive defensive play in the fourth when he reached back while sliding to catch Ross' flyball near the right-field line. ...Holland has allowed 16 homers this season. ... Both teams are off Thursday. The Red Sox then open a weekend series at the New York Yankees. The Rangers, who have won seven consecutive home series, stay home for a three-game set vs. the Chicago White Sox.

Barclays reports 9 pct gain in H1 adjusted profit

(AP) ? Barclays PLC has reported a 9 percent gain in adjusted net profit in the first half of the year thanks to lower operating costs and modest growth in income.

The bank said Friday that adjusted net profit for the six months ending June 30 was 3.1 billion pounds ($4.9 billion) compared to 3.7 billion pounds a year earlier.

Revenue net of insurance claims was up 1 percent to 15.5 billion pounds.

Adjusted pretax profit of 4.2 billion pounds beat the analysts' consensus of 3.8 billion pounds.

The adjusted figures excluded the impact of a 3 billion pounds loss charged to own credit in the first half and an own credit gain of 2.6 billion pounds a year earlier.
